چکیده
A poor anticoagulant response to activated protein C (APC r&stance) induced hy the ArglGln8g8 mutation of the Factor V gene is, in ssversl countries, the most oommon genetic defect sssootatsd with thmmbophilic disorders. Frequencies of APC resistance between 2050% have teen observed in pstients with deep venous thrombosis, SugQedinQ the importance of the Pmtein C System acttvity. Also, in other thrombotic sttusttons ss myoomtQsl infarction or thrombosis of cerebral vessels, APC resistsncs has been occasionally referred. In s previous study, we have found s prevalence of 1.5% (QS%IC 0.31 _ 4.32) for APC resistance. in the genersl portuguese Csucsstsn population. PurposeThe aim of the present study is to evstoste the prevslenco of APC resistance in one group of patients with retinal vein occlusion (FrVO) where the contribution of the Protein C system is controversial end the mls of APC resistance not yet estsbtitied. Methods: In all psttents, APC ratio (Chromogenix COATEST APC resistance M in s bioM&iewc Hemotab msgulometer) and Fsotw V gene mutation (PCR and Ynll digestion) were determined aocotiing to the reoommended fvotocols. The cut off point of 2.5 for APC ratio achieves the best agreement between phenotypic and QetIOtypiC data. We have studied 52 consecutive unrelated patieots wtth en history of RVO deftned by Hsyreh’s ortterta. Resuk: None of the observed pstients canted the mutation and the APC rstio meanrSD wss of 4.12 f 0.83 However, one patient had so APC ratio of 1,7. Conclusions: The frequencies found in the studied group sre identical to those of the general population. In this wsy APC msNtsnce. Uoes not play s major role in the possible thrombophilic component in RVO.

Demystifying EPR: A Rookie Guide to the Application of Electron Paramagnetic Resonance Spectroscopy on Biomolecules
Electron Paramagnetic Resonance (EPR) spectroscopy, also known as Electron Spin Resonance(ESR) especially among physicists, is a strong and versatile spectroscopic method forinvestigation of paramagnetic systems, i.e. systems like free radicals and most transition metalions, which have unpaired electrons. The sensitivity and selectivity of EPR are notable andintriguing as compared to other spec...
متن کاملIn vivo detection of radicals in biological reactions.
In vivo detection of free radicals on a quantitative basis is essential for an establishment of their role in membrane, protein, and cell and tissue damage. This article compares optical methods, including chemiluminescence, electron paramagnetic resonance (EPR) and NMR methods for the study of free radicals and free radical damage, especially reperfusion injury neutrophil accumulation 6 hr aft...
متن کاملElectron spin resonance and spin trapping technique provide direct evidence that edaravone prevents acute ischemia-reperfusion injury of the liver by limiting free radical-mediated tissue damage.
A novel free radical scavenger, 3-methyl-1-phenyl-2-pyrazolin-5-one (edaravone), is used for the treatment of acute ischemic stroke and is protective in several animal models of organ injury. We tested whether edaravone is protective against acute liver warm ischemia/reperfusion injury in the rat by acting as a radical scavenger. When edaravone was administered prior to ischemia and at the time...
متن کاملDirect evidence that oxygen-derived free radicals contribute to postischemic myocardial dysfunction in the intact dog.
Electron paramagnetic resonance (EPR) spectroscopy was used to investigate whether (i) the free radicals produced in the "stunned" myocardium (myocardium with postischemic contractile dysfunction) are derived from O2, (ii) inhibition of radical reactions improves function, and (iii) i.v. spin traps are effective. Open-chest dogs undergoing a 15-min coronary occlusion received an i.v. infusion o...
متن کاملIschemia-induced norepinephrine release, but not norepinephrine-derived free radicals, contributes to myocardial ischemia-reperfusion injury.
BACKGROUND Norepinephrine (NE)-derived free radicals may contribute to myocyte injury after ischemia -reperfusion, so the influence of sympathetic denervation on myocardial ischemia - reperfusion injury was investigated in the present study. METHODS AND RESULTS Cardiac sympathetic denervation was produced in Wistar rats by a solution of 10% phenol 1 week before ischemia. Atenolol (0.5 mg/kg) ...
